${it AAR3}$, a gene encoding a DCN1-like protein, regulates responses to the synthetic auxin 2,4-dichlorophenoxyacetic acid in arabidopsis roots

We have screened Arabidopsis mutants for root growth resistance to a putative antiauxin, ${it p }$-chlorophenoxyisobutyric acid (PCIB). We have successfully isolated at least three novel antiauxin-resistant mutants (${it aars}$). The ${it aar3}$, mutant was also resistant to 2,4-D as shown by a root growth assay. The ${it AAR3}$ gene encodes a protein with a domain of unknown function (DUF298). The protein has a putative nuclear localization signal and its nuclear localization was confirmed by a protoplast transient assay. The protein shares homology with the DEFECTIVE IN CULLIN NEDDYLATION-1 protein through the DUF298 domain. However, no significant difference in the auxin responsive markers, ${it DR5:GUS}$or ${it HS:AXR3NT-GUS}$, was observed between the wild type and ${it aar3}$, suggesting that the ${it AAR3}$ gene regulates 2,4-D sensitivity through a previously uncharacterized mechanism.

Root morphology and hormonal response in ${it Arabidopsis thaliana}$ grown under the continuous mechanical stimulation

We have previously proposed a new and simple growing system to dissect morphology and elongation by continuous mechanical stimulation on ${it Arabidopsis}$ roots: roots are allowed to grow on dialysis membrane-covered agar plate placed horizontally, resulting in providing continuous mechanical stimulation upon root tip during bending downward. Growth of horizontally grown ${it Arabidopsis}$ seedlings was found to be almost half as compared to that of vertical control and the reduced elongation of horizontal roots was attributed to enhancement of the ethylene response without that of the ethylene production. The role of auxin in the morphology will also be discussed based on ${it DR5:GUS}$ and ${it IAA2:GUS}$ expression study and the estimation of auxin responsive genes by real-time PCR.

Molecular characterization of flavonoid transport-related factors in flavonoid accumulating cells of Arabidopsis

Flavonoids are secondary metabolites with many important functions in plants, such as acting as pigments and UV-B protectants. Although the mechanisms by which flavonoids are synthesized in the cytosol are considerably elucidated, the mechanisms by which flavonoids are sequestered into the vacuole are poorly understood. We have isolated Arabidopsis mutants with a reduced level of flavonoid pigments, named transparent testa19 (TT19) mutants. The TT19 gene was shown to encode a protein similar to glutathione S-transferase (GST) family. We described here the characterization of flavonoid transport-related factors, such as TT19, in flavonoid accumulating cells of Arabidopsis.

Analysis on mutation induced by ion-beam and $$gamma$$-rays in ${it Arabidopsis}$

Ionizing radiation induces DNA damages which cause mutation. Organisms have DNA repair systems to repair DNA damages. The relationship between types of mutation induced by ionizing radiation and DNA repair systems are well studied in bacteria and animal. In plant, however, little is known about it. In this study we will investigate ion-beam and $$gamma$$-rays induced mutations in plant by using ${it Arabidopsis}$ mutation detection system (Yoshihara et al. 2006). Ion-beam and $$gamma$$-rays are irradiated to ${it Arabidopsis}$ containing ${it E. coli rpsL}$ gene. Mutated ${it rpsL}$ gene is collected as a plasmid by plasmid rescue from chromosomal DNA and analyze the types of mutations. We will report the results of ion-beam and $$gamma$$-rays induced mutations detected by this system.
